The action potential activity levels are regulated by chemicals called neurotransmitters, which can be either excitatory or inhibitory. Neurotransmitters are produced and secreted when their receptors are stimulated, for example when the active ingredient of a medication binds to them. Opioid receptors are found throughout the CNS, and they belong to a family of G protein-coupled receptors, which respond in the same way whether they are stimulated by opioids produced by the body, such as endorphins, or external opioids such as codeine or morphine. Codeine is metabolised into morphine after ingestion and this then binds to the opioid receptors, increasing your threshold for pain by reducing the rate of noxious (pain) action potentials along the C fibres.
Zapain tablets are quickly absorbed by the gastrointestinal (GI) tract and have a high bioavailability of 90%, which means that only 10% of the active ingredients are lost when they travel from the GI tract to the brain via the cardiovascular system. You will start to feel the effects of the medication after about 30 minutes, and it reaches peak plasma concentration after around 1 hour. The onset of action may vary slightly depending on certain factors such as your body mass index (BMI) and metabolism. The level of absorption and bioavailability, however, is not affected if you take it on an empty stomach or with food. Zapain lasts for between 4 and 6 hours, which is the amount of time that you will benefit from its pain-relieving properties. The elimination half-life is on average 3 hours, which is shorter than its duration of action. The elimination half-life refers to the time it takes for half the amount of the medication to be eliminated from the body, which means that it will be detected in your system for at least twice the amount of this time.
The exact Zapain dosage will vary according to the type and severity of your pain. Zapain 30 mg 500 mg is the most commonly used dose for moderate pain, and 1 to 2 tablets should be taken per dose, up to a maximum of 8 tablets per day. If you are taking it for chronic pain, then you should take your first dose in the morning and re-dose throughout the day as necessary. Zapain should ideally be a short-term treatment option. If you are using it for acute pain such as a migraine or period pains, then it should not be taken for more than 3 days. It is sometimes necessary to take it for a few more days or even weeks in a row if you are suffering from more severe pain and your symptoms are not going to resolve quickly, for example if you are recovering from surgery. Some people will need to take it long-term for chronic conditions such as arthritis. One reason it is not ideal as a long-term treatment option is that your body will build tolerance to the active ingredients and therefore it will become less effective over time, or you will need to increase your dose to achieve the same benefit. If you have been taking it for several weeks or months for more than 4 days per week each week, then you will need to gradually taper off the medication to reduce unpleasant withdrawal symptoms.
